Mechanism of Action
A deuterated form of tetrabenazine — substituting deuterium for hydrogen at key positions — that extends the drug's half-life, allowing less frequent dosing and more stable plasma levels with fewer peak-related side effects. Reversibly inhibits VMAT2 to reduce chorea. FDA-approved 2017 (twice-daily), with a once-daily extended-release formulation (Austedo XR) approved for both HD chorea and tardive dyskinesia.
Dosing Notes
Twice-daily Austedo titrated from 6 mg/day; once-daily Austedo XR available in 6, 12, and 24 mg strengths.
